چکیده

Borzani and Vairo (1959) reported that grampositive and gram-negative microorganisms are fundamentally different with respect to crystal violet adsorption. They claimed that the binding of crystal violet by gram-positive cells followed the Freundlich and the Langmuir adsorption laws; whereas the binding of crystal violet by gram-negative cells did not follow any known adsorption law. The present paper presents evidence which does not support their conclusion regarding gram-negative cells. It has been found that the binding of crystal violet by both grampositive and gram-negative microorganisms can be interpreted as obeying adsorption laws.

Measurement of gram-positiveness.

Several papers have been published (Kennedy and Barbaro, 1953; Barbaro and Kennedy, 1954; Bartholomew and Finkelstein, 1954; Kennedy and Woodhour, 1956; Barbaro et al., 1956) on methods of comparing degrees of gram-positiveness and the influence of certain factors on the quantity of crystal violet retained by the microorganisms. These papers compare the degrees of gram-positiveness by measuring...

Crystal violet binding capacity and the Gram reaction of bacterial cells.

Kennedy and Barbaro (1953) presented data suggesting that crystal violet adsorption values correlated directly with the gram staining character of bacterial cells, and implied that such quantitative dye uptake studies might prove a basis for a much needed quantitative index of gram positivity. Their results are consistent with a popular theory of the mechanism of gram differentiation presented ...

Quantitative Studies of Differential Staining Reactions

It has been shown previously (Kennedy and Barbaro, 1953) that the amount of crystal violet adsorbed by bacteria can be measured quantitatively by either one of two procedures. One procedure involved micro-Kjeldahl analyses of bacterial cells before and after staining. The increase in total nitrogen in the stained cells represented the dye nitrogen adsorbed. The second procedure was based on mic...

Relationship of cell wall staining to gram differentiation.

Bartholomew and Mittwer (1951) presented photographic evidence that the gram-positive staining area of bacterial cells could be fitted internally to the area demonstrated by the Dyar cell wall stain. Their conclusion was that the bacterial cell wall was not included in the grampositive area. Lamanna and Mallette (1954) explained Gram differentiation of intact cells on the basis that crystal vio...

بهینه سازی استخراج DNA ژنومی باکتریایی به روش سیلیکاژل

Background & Objectives: Today, major progress has been made in molecular experiments. The first step towards improving these experiments is the accurate extraction of nucleic acid. In this study, a protocol for DNA extraction was proposed in accordance with silica gel method. DNA purification, developed based on the silica-gel-membrane technology, is a simple method, which involves three s...
